## Abstract

BRICKET asks whether blood-test-guided encorafenib plus cetuximab retreatment benefits patients with BRAF V600E-mutated metastatic colorectal cancer after earlier benefit from the combination. Clinicians in the sources reviewed question retreatment, while support for continuing targeted therapy with a chemotherapy switch remains conditional. That alternative attracts interest; molecular selection and durability draw scrutiny. Treatment after progression remains an open research question. The sources reviewed for this edition place the trial’s main results at ESMO 2026.

## Open Medical Affairs question

What comparative evidence would justify continued EC with a chemotherapy switch, and what can BRICKET actually establish about ctDNA-guided treatment benefit?

Raised by the leading finding. The report shows what clinicians said and the sources behind it; it does not settle the question.

## Findings (titles)

- F1. Continuing EC with a chemotherapy switch attracts support, explicitly as extrapolation
- F2. The negative reading concerns EC retreatment, with the conclusion kept tentative
- F3. Insufficient molecular selection and no testing benefit are different conclusions
- F4. Durability and small-study limitations challenge positive/negative labels
- F5. Research rationale persists without a claim of demonstrated retreatment benefit

## What sits behind each finding

Counts describe what the report retained, not how many clinicians hold a view. A statement can support more than one finding.

| Finding | Statements | Passages | Sources | Named voices | Counterview | Limitations |
| --- | ---: | ---: | ---: | ---: | --- | ---: |
| F1 | 8 | 8 | 1 | 6 | recorded | 3 |
| F2 | 4 | 4 | 4 | 3 | none recorded | 3 |

## Sample findings

Paraphrased; the full report links each statement to its source.

### F4. Durability and small-study limitations challenge positive/negative labels

An X post poses a cross-study interpretation question, rather than reporting a head-to-head comparison.

### F1. Continuing EC with a chemotherapy switch attracts support, explicitly as extrapolation

An X discussion poses an analogy as a question, rather than reporting a tested comparison.

## Scope and limits

This report does not judge efficacy, estimate HCP prevalence or consensus or give treatment advice.

- Discussion that does not name the trial may be missing from this edition.
- Some clinician views in the sources used in this report are not yet captured as evidence in this edition.
- Some passages in the sources used in this report were not fully assessed for clinician views, so a view there may be missing from this edition.
- 13 statements in the sources reviewed for this edition are about other patients or topics, or only mention the trial, and are not counted.
- Further source limits apply, so some matching discussion may be missing.
- The sources reviewed for this edition are X posts and quoted or linked material, not a representative sample of HCP practice. Reposts and duplicate quotations are not additional independent opinions.
- The search emphasized the trial identifier and names; unnamed references and transcription variants may have been missed. More than 2,000 sources without the trial name were not read, and one potentially relevant discussion was not fully assessed.
- No separate protocol was listed and no BRICKET results publication was identified. Reported numerical outcomes therefore describe what authors said, not a full appraisal of trial results.
- Some speakers' identities or clinical roles are not established in the sources used in this report; each statement is attributed only as far as its source shows.
- No substantive safety or tolerability commentary was identified in the reviewed material. This does not establish an absence of toxicity or a favorable benefit–harm balance.
- Post dates do not establish presentation dates, changes of mind or disagreement across different settings.
- Cross-study PFS comparisons and biological explanations offered in the posts do not establish comparative efficacy or causal resistance mechanisms.

## Editions and corrections

- v1.0.0, accepted 2026-10-28: current edition, corrected 2026-10-29
  - Correction 2026-10-29: Corrected to match the delivered report: 10 people matched to verified clinician profiles (was 12); the sample findings.

Last checked for new commentary 2026-10-28. Versions follow semver: a new or changed finding is a minor version, more evidence without a material change is a patch.
